I glued together a room yesterday, and to make sure the floor would be nice and flat I took the trouble of sanding the tiles down using my fancy new sanding machine.  I was rewarded for my trouble by half of the tiles falling off the cork.  Does sanding the tiles down make it more difficult for the glue (white wood glue) to bond to the tiles?

Yes, plaster dust will interfere with the bond between your piece and the cork board. I wipe sanded blocks on a piece of cloth before gluing. However, IMO that's not your problem. I don't know how fancy your sanding machine is but I am surprised any of your blocks stayed glued to the cork board while using it.

Now if you glued your tiles upside down -- that is, just glued to each other not to a cork board -- waited 24 hours and then ran the piece through a really fancy thicknesser set to 1/4 inch...
